BIOENERGETICS AND AGING BIOLOGY: MITOCHONDRIA AS CROSS-FUNCTIONAL TARGETS

Abstract

Reelin/restriction point is an indication of ageing and a significant factor of cellular ageing.  This research is carried out utilizing both human and animal model of diverse ages on a mixed-method experimental design to examine mitochondrial bioenergetics.  The crucial variables that were measured using high-resolution respirometry, fluorescence assays, qPCR, and enzymatic profiling, included ATP production, membrane potential, reactive oxygen species (ROS) production, and respiratory complex activity.  The findings were that production of the ATP as well as the membrane potential decreased very much with age. Meanwhile, the production of ROS increased, and the mitochondrial complexes I and IV lost their enzymatic activity.  The copy count of mitochondrial DNA (mtDNA) and the expression levels of bioenergetic genes (ND1, COX1, ATP5B) also showed a steady reduction in their expression levels with age.  The models combining bioenergetic and molecular characteristics to train machine learning were able to predict age bracket (R 2 > 0.85), and SHAP analysis revealed that ROS and membrane potential were the most significant predictive characteristics.  These results are displayed in the form of nine tables and twelve complex visualizations. The use of expert interviews supports them because they reveal that they can be applied in different situations.  This Simon study indicates the extent to which brown mitochondaguher is deficient, and how mitochondria at once are integrable targets-Parkes of intervention therapeutic to## biological age guessing.
